Mendelovo Centrum pro vzdělání v biologii, biomedicíně a bioinformatice a Centrum molekulární biologie a genové terapie FN zvou na přednášku MicroRNA Mir-155 and Myb Proto-Oncogene Cooperate in Pathogenesis of Chronic Lymphocytic Leukemia. 10.května 2010, v 14:30 hodin posluchárna Centra molekulární biologie a genové terapie IHOK FN Brno (pracoviště Dětská nemocnice, Černopolní 9, 613 00 Brno) MUDr.Tomáš Stopka, Ph.D. Abstract: Our study involves patients (NPTS=46) with chronic B-lymphocytic leukemia (B-CLL) that all display elevation of microRNA miR-155 (compared to normal Bcells), that represents, as supported by others, novel candidate pathogenicfactor in B-CLL (Marton et al, 2008; Wang et al, 2008). All patients in this study were analyzed for prognostic markers including expression of CD38 and ZAP-70, IgVH mutation status and cytogenetics by FISH. We herein presentevidence that MYB is also overexpressed in B-CLL and positively regulates miR-155 levels. These data are supported by evidence indicating that transcription factor MYB (Myeloblastoma protein) is associated with lymphocytic development and its targets are involved in development of B-cells (Lei et al, 2004). MYB physically associates with the promoter of MIR155 host gene (MIR155HG, also known as B-cell integration cluster) in B-CLL and stimulates its transcription in reporter assay.
